Microbiologist here……..you should be plating 1 mL of water so you know how many cfu per mL or filter 100 mL using a filter cup with grid paper you can put on media to grow bacteria. Your diluted swab method is not an acceptable way to culture a water sample.

What if you used the cotton swab directly instead of diluted solution? I find it extremely hard to believe that water has zero microbial life.
